    摘要 目的:探究枕神经电刺激(occipital nerve stimulation, ONS)对慢性偏头痛(chronic migraine, CM)大鼠模型疼痛相关行为的影响。方法:采用炎症汤(inflammatory soup, IS)刺激大鼠硬脑膜模型,大鼠用随机数字表法分成慢性偏头痛模型组(CM组,n=7)、枕神经电刺激干预慢性偏头痛大鼠模型组(ONS+CM组,n=7)、对照组(Con组,n=7),对CM组和ONS+CM组连续14天输注IS刺激硬脑膜,1次/日,第15天对ONS+CM组进行ONS,基线及第1、5、9、14次IS输注后24h测量大鼠头面部多部位机械痛阈及疼痛相关行为。机械痛阈采用von Frey丝测定,疼痛相关行为采用CinePlex?行为分析系统采集大鼠15分钟的行为数据。结果:重复IS刺激硬脑膜可使得大鼠头面部机械痛阈下降并维持在较低水平,ONS可逆转这种痛阈降低;ONS可逆转因重复IS刺激引起的大鼠水平运动总距离、探索行为及冻结行为的改变。结论:枕神经电刺激可改善慢性偏头痛大鼠模型痛觉超敏现象,并可逆转部分疼痛相关行为的改变。

    Abstract:

    Objective:To examine the effects of occipital nerve stimulation(ONS)on mechanical threshold(MT) and pain-related behaviors in a rodent chronic migraine(CM) model to explore the underlying mechanism of ONS in migraine. Methods:21 Male Sprague-Dawley rats were randomly divided into Con(n=7),inflammatory soup (CM) (n=7), inflammatory soup + occipital nerve stimulation(ONS+CM)(n=7)group. A classic migraine rat model was established by repeated dural infusions of IS. The ONS+CM cohort was implanted bilaterally with electrodes to produce ONS. Periorbital、prefrontal and occipital mechanical threshold were evaluated with von Frey filaments and pain-related behaviors were tested. Results:There is no baseline differences in mechanical threshold and pain-related behaviors between the Con、CM and ONS+CM group. After the chronic migraine animal model was established, animal model displayed behavioral results similar to those experienced by migraineurs. Specifically, there was a decrease in routine physical activity and increase in Freezing episodes. Also, the animals demonstrated mechanical allodynia in the periorbital, prefrontal and occiput region. They were ameliorated by the treatment of ONS. Conclusion:Occipital nerve stimulation improves mechanical allodynia and pain-related behaviors in a rodent CM model.
